What does it mean guys down Rochester way say they're fishing at the 25th 26th or 27th.

Latitude.   If someone talks about fishing the 26 line, they are referring to 43°26' N

The other way you will hear folks communicate is with their west number (Longitude) and a depth.  So say 30.5 west and 350 feet of water for instance.  This seems more common on the east end of the lake.  Of course you have the guys who communicate via landmarks as well (The High Rocks, The Plant, 5 pipes, The Racetrack, The Radio Tower, The Sh1t plant, Front Door, Weather buoy, 2-stacks, Rudy's, The College, The Rice Bowl, The Can, West 9, the list goes on....)

Depends what port you fish out of.  At The Oak, break wall is about 43° 22.5" N.  I'm guessing Rochester is a bit south of that.
